If a Contact is manually uploaded into BrandBot and then later on they make a profile in the booking platform with the same email address, will the profiles be combined in BrandBot?

Yes, as long as the email address that was manually uploaded for the contact is the EXACT SAME as the email address that is used to create a profile within the Booking Platform, the two accounts will be combined into a singular BrandBot Contact so that purchases, reservations, etc. from the Booking Platform can be tracked on the Contact's Timeline within BrandBot.
